Evaluation of a nested-pcr for Mycobacterium tuberculosis detection in blood and urine samples BJM

The polymerase chain reaction (PCR) and its variations, such as the nested-PCR, have been described as promising techniques for rapid diagnosis of tuberculosis (TB). With the aim of evaluating the usefulness of a nested-PCR method on samples of blood and urine of patients suspected of tuberculosis we analyzed 192 clinical samples, using as a molecular target the insertion element IS6110 specific of M. tuberculosis genome. Nested-PCR method showed higher sensitivity in patients with extrapulmonary tuberculosis (47.8% and 52% in blood and urine) when compared to patients with the pulmonary form of the disease (sensitivity of 29% and 26.9% in blood and urine), regardless of the type of biological sample used. Feed restriction on performance and blood parameters of broilers fed diets with different sodium levels Rev. Bras. Ciênc. Avic.

The experiment was conducted to evaluate the effect of feed restriction and different sodium levels in the diet on the performance and hematological values (erythrocyte, hemoglobin, hematocrit, mean cell volume, mean cell hemoglobin, mean cell hemoglobin concentration and white blood cell) of broilers from 22 to 42 days of age. A completely randomized design was used in a 4 x 2 x 2 factorial (levels of sodium: 0.16%, 0.19%, 0.22% and 0.25%; with or without feed restriction; male and female), and two replicates of thirty broilers each. ABSTRACT Tambaqui,Colossoma macropomum is a fish of primary importance in Brazilian aquaculture and in the Amazon region in particular. The aim of this work is to analyze the combined effects of physical training and levels of dietary cassava (Manihot esculenta) on the hematological parameters, food intake, conversion efficiency, growth ratio and swimming performance of this fish. A diet for tambaqui consisting of 30% cassava caused decreases in weight gain and specific growth rate compared with the control group. Diets containing 15% or 45% cassava did not affect these indices and did not cause hematological changes in tambaqui juveniles, indicating that some amount of cassava can be used as an alternative energy source for this important aquaculture...

The objective of this study was to investigate the influence of dietary supplementation with propolis on hematology and number and area of melanomacrophage centers in spleen and kidney of tilapia. After acclimation, fish (24.7 ± 7.4 gmean weight) were distributed in 6 tanks 100 L, 6 fish per tank, in triplicate, at a temperature 24.0 ± 2.8oC, with two treatments: Fish fed 2% propolis supplemented diet and fish fed non-supplemented diet. The objectives of this study were to determine if protein-energy malnutrition (PEM) could affect the hematologic response to lipopolysaccharide (LPS), the interleukin-1β (IL-1β) production, leukocyte migration, and blood leukocyte expression of CD11a/CD18. Two-month-old male Swiss mice were submitted to PEM (N = 30) with a low-protein diet (14 days) containing 4% protein, compared to 20% protein in the control group (N = 30). Abstract The present study aimed to investigate the consequences of dietary levels of crude protein (CP) and total sulfur amino acids (TSAA, Met+Cys) on productive performance, egg quality and egg components for Lohmann Brown laying hens through 18-34 weeks of age. A 2×3 factorial design experiment was conducted using two levels of CP (16 and 18 %) and three levels of Met+ Cys (0.67, 0.72 and 0.77 %). A total of 144 Lohmann Brown laying hens at 18 weeks of age were randomly divided into 6 treatment groups. The best values of final body weight and body weight change were recorded by the hens fed 18 % CP diet. Intraerythrocytic organic phosphates and hemoglobins of skua - Catharacta maccormicki (Stercoraridae): at two different stages of the year in relation to Antartic migration BABT

Catharacta maccormicki blood samples were collected in the winter (October) and in the summer (February) in order to study the intraerythrocytic organic phosphates, hemoglobin (Hb) electrophoretic patterns, oxygen blood equilibrium and stripped Hbs, as well as the effect of 2,3-biphosphoglycerate (BPG) and inositol hexaphosphate (IHP) on oxygen affinity. All the samples (five from the winter and five from the summer) showed the same electrophoretic pattern: one minor fast component and one major slow one. No differences in oxygen affinity and Bohr effect in the samples collected in the winter and in the summer were found. Oxygen affinity was higher in the stripped Hb than in the blood. Light microscope observation of circulating human lymphocytes cultured in vitro BABT

The purpose of this work was to study the isolation and a light microscopy technique for cultured lymphocytes. Blood samples were obtained by venipuncture with an anticoagulant added and centrifuged in a Percoll density gradient to separate the leukocytes. Lymphocytes were placed in 25 cm ³ tissue culture flasks at 37ºC. After culturing, they were fixed and stained with the methods used for blood smears. Results showed that not all fixing solutions and stains were an equally good choice for cultured lymphocytes.

Methodological limitations of counting total leukocytes and thrombocytes in reptiles (Amazon turtle, Podocnemis expansa): an analysis and discussion Acta Amazonica

The aim of this paper is to compare three different methods for counting white blood cells [WBC] (Natt and Herrick method, estimation with 1,000 and 2,000 erythrocytes) and three methods for counting total thrombocytes [TT] (Wojtaszek method, estimation with 1,000 and 2,000 erythrocytes) in a South American freshwater turtle species, Podocnemis expansa, Schweigger 1812 (Reptilia, Pelomedusidae). Direct WBC counts using the Natt and Herrick method showed limitations, which are discussed here. The WBC and TT counts using 1,000 erythrocytes from blood smears are not recommended for Amazon turtles nor other reptilian species, since wide variation in counts can be observed. The Phrynops hilarii specie of turtle has its characterization not well defined in the literature, it was proposed in this study the leukocyte characterization of the blood, stained by Leishman and analyzed under light and transmission electron microscope. It was not observe any cellular type with similar characteristics to neutrophils in mammalian group. We believed, based on the data obtained in this study that the heterophils have a morphofuncional analogy with another neutrophils belonged to mammalian group. Abstract: This study investigated the physiological effects of a pesticide based on Bacillus thuringiensis (Dipel-WP®) added in the water and diet of Piaractus mesopotamicus during 24 and 48 h. It was added 0.13 g of de B. thuringiensis per kg of commercial feed; and for the fish subjected to the biopesticide in the water of the tanks, it was added 0.13 g/L of the biopesticide. This study was conducted to develop equations to predict chemical composition of head, limbs, hide and blood in cattle. A database containing 335 animals from 10 trials, with 221 Nellore, 38 Nellore-Simmental and 76 Nellore-Angus (96 steers, 118 heifers and 121 bulls) animals was used. Models were constructed to estimate water, ether extract (EE), crude protein (CP), ash and macrominerals (calcium, phosphorus, magnesium, sodium and potassium) in the non-carcass parts of cattle. Avaliou-se o comportamento, as respostas sanguíneas de animais confinados recebendo dieta com produto à base de própolis (LLOS). Os tratamentos foram: controle (CON) e dois produtos à base de própolis (LLOSC1) e (LLOSC1+). Mediram-se os comportamentos: comendo (COM), bebendo água (BEB), andando (AND), ruminando em pé (RUEMP), ruminando deitado (RUDET), em pé (EMP) e deitado (DET). Maior tempo em COM, BEB, AND, RUEMP e EMP ocorreu de manhã (MA) e à tarde (TA), e em RUDET e DET à noite (NO) e madrugada (MD). Animais no Lado 1 do confinamento permaneceram mais tempo andando na MA e à NO, sem diferenças para MD, e mais tempo EMP para os do Lado 1 na MA, T e MD, e igual à NO. Protective effect of sodium selenite on genotoxicity to human whole blood cultures induced by aflatoxin B1 BABT

The aim of this study was to investigate the effects of selenium and aflatoxin on human whole blood cultures (WBC) in relation to induction of sister-chromatid exchange (SCE). Results showed that the frequency of SCEs in peripheral lymphocytes was significantly increased by the direct-acting mutagen AFB1 (at doses 5 and 10 µM except for 1µM) compared with controls. When sodium selenite (Na2SeO3) was added at a molar ratio of 5x10-7 and 1x10-6, cells did not show significant increase in SCE frequency. Whereas, SCE rates induced by the various AFB1 concentrations could be significantly reduced by the presence of Na2SeO3 in a clear dose-related manner. This paper aimed at evaluating the influence of diets containing different isotopic values of carbon-13 turnover on the half-life of egg (yolk + albumen), yolk and albumen individually, and blood of poultry using delta‰13C isotopic variation. Commercial layers fed four experimental isocaloric and isonitrogenous diets (RC4, RC3, RMC4 and RMC3) containing different isotopic values, during an experimental period of 56 days. The objective of this study was to evaluate intake, apparent digestibility, ingestive behavior and blood parameters of milking goats fed cactus pear (Opuntia ficus indica) and increasing urea levels in substitution to soybean meal. Ten multiparous Alpine goats with average weight of 31±6.0 kg were used and distributed into (5 × 5) Latin squares and assessed during five 17-day periods, receiving five treatments with the following substitution levels: 0; 5; 10; 15 and 20 g/kg of urea. Neutral detergent fiber (NDF), non-fiber carbohydrates, ether extract, organic matter and water intake values showed differences, whith average values ranging from 1.03 to 1.13; 0.12 to 0.14; 1.76 to 2.10 kg/day and 0.77 to 1.48 kg/day, respectively.
